Ferritin, a major iron storage protein found in most living organisms, is composed of a 24-subunit protein cage and an inner iron oxide core.Based on the unique structure of human H-ferritin (HFn), we biomimetically synthesized a new kind of ferritin nanoparticle, named magnetoferritin (M-HFn).Then, we proved the HFn protein shell of M-HFn could specifically target to tumor cells via binding its receptor TfR1, and we further demonstrated that Fe3O4 core of M-HFn exhibits the peroxidase activity that visualizes the tumors in the presence of substrates.Based on this bifunctional nanoparticle, we developed a new method for cancer diagnosis.After screening 541 clinical specimens of 10 kinds of human cancer tissues by this method, we verified that it can distinguish cancerous cells from normal cells with a sensitivity of 95.77% and specificity of 95.72%.
