As a traditional Chinese medincine,Ramulus mori has lots of active compounds eg.flavonoids,stilbennoids etc [1-2].In order to separate and purify them from each other,former researchers generally chose the column chromatography [1-2],while it is too complicated and inefficiency for the separation process.In this paper,high-speed counter-current chromatography (HSCCC) [3] is applied to the purification of these components,andhigh-purity components can be obtained from Ramulus mori with this technology.HSCCC is a kind of continuous liquid-liquid distribution/partition chromatography with the unique feature of eliminating the use of a solid support,which subsequently eliminates all complications arising there from,such as absorptive loss and denaturation of samples.Here,its procedure was performed in a two-phase solvent system (Arizona solvents system) where the operational condition is shown as follows:the proportion of the solvent is n-hexane:ethyl acetate:methyl:water =1∶1∶1∶2,at the normal elution mode,the flow-rate,rotation speed is set at 2.0mL/min,900rpm respectively and 10ml of sample solution containing 75mg of the crude extraction is injected for the purification process.According to this process,four high-purity compounds is obtained in 180 min,Oxyresveratro 1 (25mg,97%),trans-resveratrol(7mg,95%),Moracin M(29mg,97%)and cis-resveratrol(3mg,95%),All of them are identified by HPLC,MS,1H-NMR and 13C-NMR,.As the results,it indicates that HSCCC is an efficient and effective way for the separation of the active compounds in Ramulus mori.,and it is shown many advantages compared with the column chromatography method.
