Apolipoprotein C-Ⅲ (apoC-Ⅲ) is a small, interchangeable apolipoprotein present in HDL, VLDL and to a much smaller extent in LDL.We have shown that apoC-Ⅲ is a strong inhibitor of the clearance ofVLDL, IDL and LDL in vivo in humans.The result of the inhibition of VLDL and IDL clearance is a prolongation of their intravascular halflife, which leads to the formation of small, dense LDL.Besides its deleterious effects on lipoprotein metabolism, apoC-Ⅲ has several direct proatherogenic properties on the vascular wall, increasing expression of adhesion molecules by endothelial cells and monocytes, and inducing a dysfunctional, insulin-resistant phenotype in endothelial cells.
