Porous silica nanoparticles (PSN) are universally addressed as a major candidate for use as carriers in controlled drug delivery systems. However, most templates used to prepare PSN are poisonous surfactants. To reduce the toxicity and improve stimulatory of surfactants, the surfaetant templates must be removed from the pore channel systems before drug molecules were loaded into PSN in the traditional preparing process. In fact, there is no guarantee that the surfactants will be removed completely from the pore channel systems, and this loading method (drug loading after PSN formation) is far inferior to in situ loading (drug loading and PSN formation by one step) in drug loading ability.
