Background Linkage has been reported between allelic variations in HLA-DR and Parkinsons disease (PD), but the polymorphisms were found to be different ethnically and geologically.HLA-DR and PSMB, TAP loci are all located in the HLA class Ⅱ region, and they play important roles in both immune response and protein degradation in neurodegenerative diseases.To date, little research has been performed to investigate the association between PD and polymorphisms of HLA-DRA, PSMB and TAP in Han Chinese.To investigate the association between the polymorphism of HLA class Ⅱ region and sporadic PD in Han Chinese population, a case-control study was performed through genotyping single nucleotide polymorphisms (SNPs) in HLA-DRA, PSMB8, PSMB9, TAP1, and TAP2 genes.

人白细胞抗原(HLA)参与了PD的发生、发展.HLA-DR、PSMB区基因相邻,当染色质处于开放构象状态时,这些基因在细胞的抗原呈递过程中或共同发挥作用.2010年欧洲GWAS证实:HLA-DRA第1内含子rs3129882与PD相关,但未获我国证实,PSMB基因编码区SNPs可影响AD患者脑细胞的免疫蛋白酶体活性,推测HLA-DRB可能参与机体的抗原呈递,而PSMB变异则影响蛋白酶体的降解功能.
