The c-Jun N-terminal kinase (JNK), also called stress-activated protein kinase (SAPK), belongs to the mitogen-activated protein kinase (MAPK) superfamily.JNK plays a crucial role in cell proliferation, differentiation, stress responses,and apoptosis.JNK kinase activity can be activated by diverse stimuli, including growth factors, cytokines, environmental stresses, and apoptotic stimuli.Activated JNK can be dephosphorylated and inactivated by various dual-specificity MAPK phosphatases (MKPs).Interestingly, we have identified two protein phosphatases, JKAP (JNK pathway-associated phosphatase) and PP4 (protein phosphatase 4), which can stimulate JNK activity.PP4 is a PP2A-related serine/threonine protein phosphatase.PP4 also binds to and modulate c-Rel/RelA, IRS-4 and HDAC3.Conditional gene targeting and proteomic approaches are being used to further characterize the signaling pathways mediated by PP4.
