Nebivolol is a novel,highly selective β1 adrenergic blocker currently used in clinical practice.However,there is less information regarding its effects on renal microcirculation.The aim of the present study is to investigate the vasoactive properties and action mechanisms of nebivolol on afferent arterioles ofjuxtamedullary nephron.We used the in vitro blood-perfused juxtamedullary nephron technique combined with videomicroscopy.Single afferent arterioles of Sprague-Dawley rat were visualized and superfused with solutions containing nebivolol or L-NNA,a nitric oxide synthase (NOS) inhibitor.In response to superfusion with nebivolol at concentrations of 0.1,1,10 and 100μmol/L,afferent arteriolar diameter increased significantly from 16.8±0.3 to 17.5±0.3,18.5±0.3,19.6±0.7 and 20.1±0.7μm (20.0±3.6%,p<0.01 via baseline).However,this effect was markedly blocked by pretreatment with L-NNA.Adding L-NNA (100μmol/L) decreased afferent arteriolar diameter significantly from 17.5±0.3 to 14.8±0.3 μm (-15.5± 1.5%,P<0.01).In the presence of L-NNA,superfusion with nebivolol at concentrations of 1,10 and 100 μmol/L,afferent arteriolar diameter increased significantly to 15.4±0.3,16.1 ±0.3 and 17.2±0.4μm (16.1 ±2.8%,p<0.01 via L-NNA group;-1.9±0.7%,p>0.05 via baseline).These results indicate that β1 adrenergic receptors are functionally expressed in juxtamedullary afferent arterioles and nebivolol predominantly activates β1 adrenergic receptors leading to vasodilation via NO mechanisms.
