Abrstract:Peroxidase (POD) activity was visualized at the cellular level of a young and mature tree of Metroxylon sagu employing histochemistry technique.Intense POD activity was observed in the cell wall,peroxysomes and to some extent in the cytoplasm and the vacuole when visualized under light microscope.The enzyme was further separated and purified using a combination of temperature-induced phase partitioning,cation-and anion-exchangers,and gel filtration.Two (2) isoenzymes namely mPOD-Ⅰ and mPOD-Ⅱ and were successfully purified to homogeneity and their biochemical and kinetic properties were characterized.
