Adeno-associated virus (AAV) vector has been successfully utilized as a therapeutic gene delivery vector in several human clinical trials.To optimize AAV vector for effective tumor gene therapy, several approaches have been explored including tumor specific targeting and enhanced therapeutic gene expression.Based on the high homology among the capsid genes from 12 independent AAV serotypes, we have utilized a DNA shuffling ofAAV serotype capsid sequences to generate a library of AAV mutants.After transfection of AAV shuffling library into Chinese hamster melanoma CS1 cells, we recovered a unique AAV variant that demonstrated remarkably high efficiency to transduce CS1 cells when compared with the parental AAV serotypes.
