6-磷酸海藻糖合成酶(Trehalose-6-phosphate Synthase,TPS)是昆虫海藻糖合成途径中的关键酶,在某些昆虫的滞育过程中起到重要作用.本文根据葱蝇(Delia antiqua)差减杂交文库中的EST序列信息,设计特异性引物并通过RACE技术克隆了葱蝇6-磷酸海藻糖合成酶基因eDNA,其全长为2904 bp(命名为DaTPS1),开放阅读框2448 bp,编码815个氨基酸,推测其相对分子质量为91.2 kD,等电点为5.96.通过该基因推导的氨基酸序列与其它物种的TPS相似性比较和系统发育分析,发现葱蝇与黑腹果蝇(Drosophila melanogaster)的氨基酸序列相似性高达92.1%.利用半定量RT-PCR方法分析发现,DaTPS1在夏滞育和冬滞育前期表达较高,滞育期较低,滞育期结束后又有所升高,由此推测TPS1可能在葱蝇滞育过程中起到重要作用.
