Over several decades,there have been continued efforts in biosensors for the sensitive and selective detection of disease biomarkers,environmental toxins and bio-warfare agents,etc.[1,2] However,the satisfied performance of these current biosensors usually suffers from the multiple operations (for example,multiple reagents delivery,separation steps,washing steps and signal amplifications) which hamper the further practical applications of these biosensors.Thus,the development of rapid,portable,highly integrated,and user-friendly point-of-care (POC) diagnostic devices is highly desirable in both developed and resource-limited countries.[3] Herein,we have designed an intelligent electrochemical microbiochip device (IEMD) with totally integrated automatic reagents delivery system and portable electrochemical detection platform which greatly reduces the operations of detection from multiple steps to single step.And thus,the personal intervention is minimized and the overall detection process is within 30 minutes.The design of meandering channel ensures the smooth and stable reagents transportation.Using this novel IEMD,we have demonstrated the sensitive and selective detection of tumor biomarkers (PSA and AFP) and mimicking virus of influenza (H1N1-SV).More importantly,we have successfully demonstrated the simultaneous detection of tumor biomarkers (AFP and PSA) to the level of pg/ml and achieved selective detection in diluted serum,and even conducted PSA assay directly in patients serum in less than 30 min.Our IEMD would be an ideal and robust platform for tumor screening and bedside assay.
