Aim.To investigate serum and urine metabonomics after carbon tetrachloride(CCl4)-induced by the liver fibrosis using gas chromatography-mass spectrometry(GC/MS).Methods.Rats induced by subcutaneous injection CCl4 twice each week for 12 weeks continuously(1.0 mL/kg 50%CCl4,diluted in olive oil).Histopathological change is to judge the success of CCl4-induced liver fibrosis model.Serum and urine samples from two groups(control and model)were collected at 12W,and the metabolic profile changes were analyzed by GC/MS.Results.Metabolic profile studies indicated that different clustering of the two groups could be separated,and seven metabolites in serum were found,such as isoleucine,carnitine,glucose,et al; five significantly changed metabolites in urine including cytidine,corticoid,et al were found.Conclusions.The pathogenesis of liver fibrosis might be related to dysfunction of energy metabolism,glucose metabolism,amino acid metabolism,P450 metabolism,fatty acid metabolism,glutathione biosynthesis metabolism,nucleic acid metabolism and water-electrolyte metabolism,which hints that through potential biomarkers for the research methods it is achievable to get new targets and ideas of innovation in new drug to cure liver fibrosis.
