纳米混悬剂可增大难溶性药物的溶解度和生物利用度,提高药物的安全性和有效性.纳米混悬剂作为一种新的给药系统,在药物中的应用受到广泛关注.本文对纳米混悬剂的制备方法进行了阐述,并对其优势存在的问题进行了分析和展望.提高溶解度和生物利用度是难溶性药物研究中的一项重要课题,也是药剂现代化亟待解决的问题.运用现代制药技术并根据不同辅料的不同性质,可以将难溶性药物依据临床不同用途而制成不同剂型,从而提高难溶性药物的溶解度和生物利用度.虽然可以通过制成脂质体、固体分散体、环糊精包合物等方法来提高难溶性药物的溶解度,但是这些方法普遍存在着载药量低、制备工艺复杂以及稳定性较差等问题.近年来国内外研究者对混悬剂给予了高度重视,尤其是纳米混悬剂(nanosuspensions).普通混悬剂中药物微粒的粒径一般在0.5~10μm之间,小者可为0.1μm,大者可达50μm甚至更大.而纳米混悬剂中药物的平均粒径小于1μm,一般在200~500nm之间.
