目的:类风湿性关节炎(rheumatoid arthritis,RA)是一种常见的累及滑膜和关节的慢性自身免疫性疾病,近年来的研究提示RA的发病与感染、遗传和内分泌等有关,但其发病机制至今仍不明确.NK细胞是天然免疫系统重要的效应细胞,同时能够调节获得性免疫,参与机体免疫病理损伤,与多种免疫疾病的发生和发展密切相关.雷公藤是我国传统中药,在多种自身免疫性疾病的治疗中疗效显著.雷公藤内酯醇(triptolide,TP)是雷公藤组份中免疫抑制作用最强的单体,研究表明TP能够调控多种免疫细胞(如T细胞、树突状细胞)的免疫功能,课题组前期工作也发现TP能够明显抑制肾脏小管上皮细胞(非经典APC)表面C3、CD40、B7h表达水平,降低炎症和免疫应答的发生,但有关TP对RA患者NK细胞功能的影响尚未见报道.本研究通过体外试验,探讨TP对RA初发患者NK细胞免疫功能的影响,为RA的发病机制及TP在RA治疗中的应用提供进一步的理论依据.
