To prevent post-exposure prophylaxis of HBV infection, we generate a fully human antibodyXy-2.The variable regions of the antibody (VH and VL) was cloned from a phage antibody library specific for HbsAg.The whole antibody genes were constructed by cloning of VH and VL genes into a vector that contains the constant regions of human IgG 1 and κ chain, and were expressed in CHO/DHFR-cells.The antibody production was improved by ZnSO4 or CdCl2 as to 50 μg/106 cells/ 24h.The antibody was purified with a protein A-sepharose column.The epitope specificity of the antibody Xy-2 was identified as PreS2 and no cross-reaction with human tissues.The in vitro neutralization assay showed that antibodyXy-2 could specifically block HBV infecting host cells and its neutralizing activity is 100 times higher than that of commercial polyclonal HBIG.Our data indicate that antibodyXy-2 has a great potential application for prevention of HBV infection.
