目的:研究芍药内酯苷对环磷酰胺致血虚小鼠的补血作用及机制。方法:以四物颗粒及芍药苷为对照,检测外周血象,胸腺与脾脏指数,骨髓病理切片,酶免法检测血浆粒细胞集落刺激因子(G-CSF)、放免法检测血清粒细胞-巨噬细胞集落刺激因子(CM-CSF)、肿瘤坏死因子-α(TNF-α)含量变化;RT-q PCR法检测脾脏中G-CSF、CM-CSF及TNF-α的基因表达;Western Blot法检测骨髓中G-CSF、CM-CSF及TNF-α的蛋白表达。结果:结果:与模型组比较,芍药苷组与芍药内酯苷高剂量组均能明显升高白细胞数和胸腺指数(P<0.01,P<0.05),升高G-CSF与GM-CSF含量(P<0.01,P<0.05),降低血清中TNF-α水平(P<0.05,P<0.01),降低脾脏TNF-αm RNA的表达(P<0.05)。病理切片结果显示,芍药苷组与芍药内酯苷高剂量组小鼠骨髓细胞数量多,密度大,巨核细胞数量与模型组相比较少。结论:芍药内酯苷具有补血作用,不仅能够通过促进造血生长因子的分泌调节造血细胞的增殖分化,还能够调控负向造血因子,减轻其对造血功能的抑制。
Objective: To study the effect and mechanism of albiflorin on blood-invigoration induced by cyclophosphamide in blood deficiency mice. Results: Compared with the model group, both the paeoniflorin group and the paeoniflorin high-dose group could significantly increase the number of leukocytes and the thymus index (P <0.01, P <0.05), and increase the contents of G-CSF and GM- (P <0.01, P <0.05), decreased the level of TNF-α in serum (P <0.05, P <0.01), and decreased the expression of TNF-αmRNA in the spleen (P <0.05). The results of pathology showed that the paeoniflorin group and the paeoniflorin high-dose group had more bone marrow cells, larger density and fewer megakaryocytes compared with the model group. Conclusion: Paeoniflorin has the function of blood enrichment, which not only can regulate the proliferation and differentiation of hematopoietic cells by promoting the secretion of hematopoietic growth factors, but also regulate the negative hematopoietic factors and reduce the inhibition of hematopoietic function.
