RNA干扰(RNA interference),是一种能够有效抑制基因表达的靶向性转录后基因沉默机制.通过这种机制,可以实现对相关疾病的治疗1.但是截止2013年上旬,依然没有一个药物能够通过FDA的批准进入药物市场,其主要原因是缺乏有效的siRNA的给药体系.

我们课题组致力于siRNA的成药性研究.我们的研究目标:一是合成修饰性siRNA从而提高血清稳定性,在一定程度上降低脱靶效应;二是对于RNA干扰通路中关键蛋白的机制研究.已经有文献证明,人体RNA解旋酶(RHA)能够将双链siRNA负载到RISC复合物上.
RNAi(RNA干扰)是一种有效的基因调控机制,它是通过依赖基因序列的方式来抑制靶基因的表达[1].基因治疗的成功在很大程度上取决于传递载体,低传递效率以及脱靶效应是目前siRNA应用于临床治疗的主要障碍,最近的研究发现,纳米颗粒可以有效地包裹RNA,并高效地将RNA传递到肿瘤细胞和其他含有网状内皮烯酮的组织中[2].
