研究表明microRNA参与调控动物原肠运动,但是microRNA调控动物原肠发生的分子机制还不清楚.为研究microRNA在小鼠原肠发生过程中的功能,我们通过microarray分析筛选到30个microRNA在小鼠E7.5胚胎三陪层中表达变化倍数在3倍以上的microRNA,其中miR-127在ES细胞分化过程中表达量明显增加.在小鼠E7.5胚胎中,miR-127主要表达在原条和内胚层.miR-127过表达不影响ES细胞干性,但是过表达的ES细胞系在分化过程中原条标记分子Foxa2和T的表达量明显增加,并且在斑马鱼体内过表达miR-127明显促进胚盾区(相当于小鼠原条)标记分子Chd和Gsc的表达.在ES细胞和EpiSC细胞中分别敲降miR-127,原条标记分子Foxa2和T则表达量下降.这些结果表明miR-127在原条发育过程中具有重要功能.进一步研究表明Lefty2是miR-127直接调控的靶基因之一,在ES细胞分化过程中过表达miR-127抑制Lefty2表达,同时促进Smad2的磷酸化.我们的研究结果表明miR-127在动物原肠发生过程中,通过Lefty2-与Nodal信号通路调节原条形成.
