目的探讨调心方对脑缺血再灌注损伤小鼠脑组织中过氧化氢酶(CAT),丙二醛(MDA)和脂褐素(LF)含量的影响。方法采用改进的Himori法制备小鼠脑缺血再灌注损伤模型。小鼠随机分为7组:正常组、假手术组,模型组、调心方40,20,10 g.kg-1剂量组、脑复康阳性对照组,连续ig给药15 d后,断头处死小鼠,取脑组织,分别采用钼酸铵显色法、硫代巴比妥酸法以及荧光分光光度法测定小鼠脑组织中CAT,MDA和LF含量。结果调心方三个剂量组均可增强小鼠脑组织中CAT活性,降低MDA,LF含量。结论调心方可通过增强CAT活性,降低MDA,LF含量改善脑缺血再灌注造成的损伤。
Objective To investigate the effects of Heart Benefiting Recipe (QIAOXIN) on catalase (CAT), malondialdehyde (MDA) and lipofuscin (LF) content in brain tissue of mice with cerebral ischemia-reperfusion injury. Methods The mouse model of cerebral ischemia-reperfusion injury was established by modified Himori method. The mice were randomly divided into 7 groups: normal group, sham-operation group, model group, 40,20,10 g.kg-1 group and Jifukang positive control group. After 15 days of continuous ig administration, The mice were killed and the brain tissue was taken out. The content of CAT, MDA and LF in brain tissue were determined by ammonium molybdate chromogenic assay, thiobarbituric acid assay and fluorescence spectrophotometry respectively. Results Tiaoxin Fang three dose groups can enhance the CAT activity in mouse brain tissue, reduce MDA, LF content. Conclusion Tiaoxin can improve the activity of CAT and reduce the content of MDA and LF to improve the damage caused by cerebral ischemia and reperfusion.
