Although callus formation is usually perceived to be a consequence of auxin-to-cytokinin gradients that are inadequate for the formation of shoots or roots, callogenesis may require additional conditions. Here, we used a collection of tomato mutants and transgenic plants in a single genetic background (cv.Micro-Tom) to elucidate the role of ethylene in callus formation. The ethylene insensitive mutant, Never ripe (Nr), failed to form callus on an improved callus-inducing medium (CIM) supplemented with 1.0 M 2,4-D and 0.5 M BAP.
