Human beings are inevitably exposed to ubiquitous phthalates(PEs)surroundings,and simultaneously ingesting a plenty of food emulsifiers via daily diet.The purposes of this study were to explore whether and how food emulsifier glycerin monostearate(GMS)could exacerbate the male reprodcutive toxicity of the mixture of six priority controlled phthalate esters(MIXPs).Male rats were orally administered either sodium carboxymethyl cellulose as controls or MIXPs at 16 mg/kg/d with or without GMS by oral gavage for 15 weeks.Testosterone and luteinizing hormone were analyzed,and histopathological examination of testis were performed for the toxicity evaluation; Steroidogenic proteins(StAR,P450scc,CYP17A1,17β-HSD and Arom),tight junction related proteins(claudin and ZO-1)and P-glycoprotein(P-gp)were measured for mechanisms exploration.Results showed that GMS exacerbated the reproductive toxicity of MIXPs,including the decrease of body weight,testicular and serum testosterone level,and the constructional damage of testis.GMS down-regulated the expression of StAR,P450scc,CYP17A1,17β-HSD,claudin and ZO-1,while up-regulated the expression of Arom in testis.The results indicated that GMS might damaged the structure and function of tight junction and P-gp efflux system,then enhanced the permeability of PEs and increased their toxicity in rats.
