通过远缘杂交,将抗灰霉病的多毛番茄PI134417单株和园艺性状优良的轮回亲本99165-30杂交,再利用F1代单株和轮回亲本回交,获得了用于构建图谱的BC1分离群体,设计染色体铆定SSR

通过远缘杂交,将抗灰霉病的多毛番茄PI134417单株和园艺性状优良的轮回亲本99165-30杂交,再利用F1代单株和轮回亲本回交,获得了用于构建图谱的BC1分离群体,设计染色体铆定SSR引物,利用SSR技术将已经用AFLP技术构建连锁遗传图定位到染色体上,构建高饱和遗传图谱。将BC1单株自交得到BC1S1自交株系,对BC1S1株系进行人工接种抗病性鉴定,并在此基础上对番茄灰霉病抗性基因进行了QTL的定位。主要研究结果如下:(1)利用染色体铆定的23个SSR标记,对本课题组已经构建的含有178个AFLP标记的遗传连锁群进行完善,构建了一张定位于染色体上的占整个基因组80%的遗传连锁图谱;(2)定位了与番茄叶抗灰霉病相关的位点2个,一个位于番茄第2条染色体上,为抗病QTL,与E40/M59-9标记紧密连锁;另外一个QTL位于番茄第3条染色体的末端,为感病QTL,与SSR11紧密连锁;(3)定位了与番茄果实抗灰霉病相关的位点1个,位于第2条染色体上,为抗病QTL,侧翼标记为E32/M59-6和SSR57。
