目的观察沙巴棕提取物与人肺癌细胞增殖和凋亡之间的关系,并探索其作用机制。方法应用MTT比色法、流式细胞仪法观察人类肺癌细胞成活、凋亡情况。应用Western blot法观察PARP、CASP3、P27、CDKN1B、p-STAT3蛋白的表达情况。结果沙巴棕提取物以剂量依赖性的方式抑制了肺癌细胞的增殖,并通过上调PARP、CASP3、P27等凋亡相关蛋白促进凋亡。沙巴棕提取物降低了p-STAT3水平;STAT3磷酸化的一种选择性抑制剂———AG490,同样通过降低p-STAT3的水平而抑制了肺癌细胞的生长。相比于以上二者任意一种的单独治疗,沙巴棕提取物和AG490的联合治疗显著降低了肿瘤细胞的增殖(P<0.05)。结论沙巴棕提取物可通过促进凋亡信号和下调p-STAT3蛋白而抑制肺癌细胞增殖。这些观察可能为沙巴棕提取物诱导肿瘤生长抑制的机制研究提供有价值的参考。
Objective To observe the relationship between the extract of Sabahan and the proliferation and apoptosis of human lung cancer cells and to explore the mechanism of action. Methods MTT assay and flow cytometry were used to observe the survival and apoptosis of human lung cancer cells. Western blot was used to observe the expression of PARP, CASP3, P27, CDKN1B and p-STAT3 protein. Results Sabouraud extract inhibited lung cancer cell proliferation in a dose-dependent manner and promoted apoptosis by up-regulating apoptosis-related proteins such as PARP, CASP3 and P27. Sabouraud extract reduced p-STAT3 levels; AG490, a selective inhibitor of STAT3 phosphorylation, also inhibited the growth of lung cancer cells by decreasing the level of p-STAT3. The combination of sababa brown extract and AG490 significantly reduced the proliferation of tumor cells (P <0.05) compared to either treatment alone or in combination. Conclusion Sabah brown extract can inhibit lung cancer cell proliferation by promoting apoptosis signal and down-regulating p-STAT3 protein. These observations may provide a valuable reference for the research on the mechanism of the inhibition of tumor growth induced by Sabah brown extract.
