目的探讨醒脑静注射液(XNJJ)对蛛网膜下腔出血(SAH)后海马相关因子凋亡的影响。方法选择成年雄性Wistar大鼠35只,随机分为空白对照组、生理盐水对照组、SAH组、XNJJ治疗1组、XNJJ治疗2组,每组7只,采用新鲜自体股动脉血枕大池二次注入法建立SAH模型,用Real-Time PCR技术检测海马神经元中凋亡分子Caspase-3和Bax以及抗凋亡分子Bcl-2在各组的变化情况,并用Western blot进一步验证此变化。结果 SAH后海马神经元中Caspase-3和Bax的表达均有不同程度上升(P<0.05),Bcl-2也有所上升(P<0.01)。XNJJ治疗1组及2组Caspase-3和Bax的表达较SAH组下降(P<0.01),而Bcl-2变化无统计学差异(P>0.05)。结论 XNJJ可以抑制海马神经元凋亡,具有脑保护作用。
Objective To investigate the effect of xingnaojing injection (XNJJ) on the apoptosis of hippocampus related factors after subarachnoid hemorrhage (SAH). Methods Thirty-five adult male Wistar rats were randomly divided into blank control group, saline control group, SAH group, XNJJ treatment group 1 and XNJJ treatment group 2, with 7 rats in each group. SAH model was established by injection method. The changes of apoptotic molecules Caspase-3 and Bax and anti-apoptotic molecule Bcl-2 in hippocampal neurons were detected by Real-Time PCR. The changes were further verified by Western blot. Results The expression of Caspase-3 and Bax increased in hippocampal neurons after SAH (P <0.05) and Bcl-2 increased (P <0.01). Compared with SAH group, the expression of Caspase-3 and Bax in XNJJ group 1 and 2 group was lower than that in SAH group (P <0.01), but no significant difference was found in Bcl-2 (P> 0.05). Conclusion XNJJ can inhibit the apoptosis of hippocampal neurons and protect the brain.
