目的探讨人脑胶质瘤干祖细胞-绿色荧光裸小鼠皮下移植瘤模型的建立。方法将人脑胶质瘤干祖细胞株SU3接种于NC-C57BL/6J荧光裸小鼠右前肢腋下,定期测肿瘤大小,取移植瘤组织块持续传代,并做常规病理、免疫组化染色和绿色荧光蛋白(GFP)检测。结果移植瘤连续传至10代,100%致瘤率。肿瘤晚期都有肉眼可见的肿瘤表面皮肤溃疡性结痂,常规病理符合恶性肿瘤特征,分子免疫病理缺氧诱导因子1α(HIF-1α)、CD34、TNF、Ki67非均一性表达于相关区域或相关细胞,在470nm激发光下可见移植瘤间质存在一定数量的GFP细胞,以及在坏死区域内的GFP细胞CD68+和CD11b+。结论成功建立人脑胶质瘤干祖细胞-绿色荧光裸小鼠皮下移植瘤模型。
Objective To investigate the establishment of human glioma stem-progenitor cells - green fluorescent nude mice subcutaneous xenograft model. Methods The human glioma stem cell line SU3 was inoculated into the axillary of the right forelimb of NC-C57BL / 6J fluorescent nude mice. The size of the tumor was measured regularly.
