目的:探讨胃癌组织中环状RNA矮小相关转录因子1(circRUNX1)的表达及其与临床病理特征和患者预后的关系。方法:收集上海交通大学附属第一人民医院胃肠外科2015年4月至2018年12月接受首次胃癌根治术的35例胃癌患者的胃癌组织(研究组)及癌旁正常组织(对照组),利用实时定量反转录聚合酶链反应(RT-qPCR)检测组织中circRUNX1表达水平。分析circRUNX1的表达水平与胃癌患者临床病理特征及患者预后的关系。统计学方法采用n χ2检验和Kaplan-Meier法结合Log-rank检验进行。n 结果:结合circRNA测序结果和RT-qPCR验证,发现胃癌组织中circRUNX1的表达水平高于癌旁正常组织(68.6%,24比11)。进一步分析发现胃癌组织中circRUNX1的表达水平与肿瘤大小(n χ2=5.844,n P<0.05)、淋巴结转移(n χ2=6.496,n P<0.05)及肿瘤病理分期(TNM分期)(n χ2=8.426,n P<0.05)显著相关。Kaplan-Meier分析结果显示,circRUNX1高表达的胃癌患者的总生存率低于circRUNX1低表达者(Log-rank=11.010,n P<0.05),差异有统计学意义。n 结论:circRUNX1在胃癌组织中高表达,与胃癌的临床病理特征和患者预后密切相关,可能成为一个潜在的胃癌预后新指标。“,”Objective:To explore the expression of circular RNA hsa_circ_0002360 (named circRNA runt-related transcription factor 1, circRUNX1) in gastric cancer and the relationship between circRUNX1 expression and clinicopathological characteristics of gastric cancer and prognosis of patients with gastric cancer.Methods:From April 2015 to December 2018, the gastric cancer tissues (study group) and adjacent normal tissues (control group) of 35 patients with gastric cancer who underwent the first radical gastric cancer surgery in the Department of Gastrointestinal Surgery, Shanghai General Hospital of Shanghai Jiaotong University, were collected. The real-time quantitative reverse transcription polymerase chain reaction (RT-qPCR) was used to detect the expression level of circRUNX1 in the tissues. The relationship between the expression of circRUNX1 and the clinicopathological characteristics of gastric cancer tissues and prognosis of patients with gastric cancer was analyzed. The n χ2 test and Kaplan-Meier combined with Log-rank analysis were used.n Results:The results of circRNA sequencing and RT-qPCR showed that the expression of circRUNX1 in gastric cancer tissues was higher than that in adjacent normal tissues (68.6%, 24 vs. 11). Further analysis revealed that the expression level of circRUNX1 in gastric cancer tissues was significantly correlated with tumor size (n χ2=5.844, n P<0.05), lymph node metastasis (n χ2=6.496, n P<0.05) and tumor node metastasis (TNM) stage (n χ2=8.426, n P<0.05). Kaplan-Meier analysis showed that the overall survival of gastric cancer patients with high expression of circRUNX1 was shorter than in those with low expression of circRUNX1 (Log-rank=11.010,n P<0.05).n Conclusion:The high level of circRUNX1 in gastric cancer tissues was associated with the clinicopathological characteristics of gastric cancer and prognosis of patients with gastric cancer, and may be used as a potential new indicator for prognosis of gastric cancer.
