目的研究从猫豆中提取的单体成分猫豆胍(MDG)的镇静催眠、抗惊厥和抗震颤麻痹作用,为猫豆的综合利用奠定基础。方法采用小鼠自主活动法、戊巴比妥钠阈下剂量催眠法、延长戊巴比妥钠睡眠时间法、抗硝酸士的宁致惊厥试验法和氧化震颤素致小鼠震颤试验法,观察MDG的镇静催眠、抗惊厥和抗震颤麻痹作用。结果在375mg/kg剂量下MDG能减少小鼠自主活动次数和活动路程(P<0.05);500、625mg/kgMDG能增加阈下剂量戊巴比妥钠入睡小鼠只数(P<0.05、0.01);625mg/kgMDG能延长戊巴比妥钠睡眠时间(P<0.05)和显著延长小鼠发生惊厥的潜伏期和死亡时间(P<0.01);MDG在375和625mg/kg剂量时,对小鼠震颤潜伏期和持续时间均无显著影响(P>0.05)。结论猫豆中提取的MDG具有一定的镇静催眠和抗惊厥作用,但没有抗震颤麻痹作用。
Objective To study the sedative, hypnotic, anticonvulsant and anti-parkinsonism effects of monomeric constituents of cat’s bean extract (MDG) extracted from cats and beans, and to lay a foundation for the comprehensive utilization of cats and beans. METHODS: The mouse spontaneous activity method, pentobarbital sodium sublimation dose hypnosis method, prolongation of pentobarbital sodium sleep time method, anti-nitric acid-induced seizure test method, and oxotremorine-induced mouse tremor test method were used. MDG’s sedative-hypnotic, anticonvulsant and anti-paralysis properties. RESULTS: At the dose of 375 mg/kg, MDG reduced the number of spontaneous activities and the distance of movement (P<0.05); 500 and 625 mg/kg of MDG increased the number of mice that fell in the subthreshold dose of pentobarbital (P<0.05, 0.01). 625 mg/kg of MDG prolonged the sleep time of pentobarbital sodium (P<0.05) and significantly prolonged the latency and death time of convulsions in mice (P<0.01); MDG at 375 and 625 mg/kg doses of mice There was no significant influence on latency and duration of tremor (P>0.05). Conclusion MDG extracted from catpea has certain sedative, hypnotic and anticonvulsant effects, but it does not have antiparkinsonian effects.
