目的分离筛选具有抗菌活性及抗肿瘤活性的药用植物内生菌。方法采集贵州地道药用植物大黄、刺梨、杜仲、白芍和刺五加等5种组织材料,经表面消毒、无菌匀浆后,用改良的GYM培养基和TWYE培养基进行分离,利用耐药大肠杆菌、耐药金黄色葡萄球菌、藤黄微球菌及白色链球菌等测试菌和稻瘟霉模型对分离菌株的发酵产物进行筛选,并通过HPLC对活性菌株发酵产物做进一步分析。结果分离得到植物内生菌180株,其中46株有不同程度的抗菌活性,占分离菌株总数的25.6%,58株菌有不同程度的抗稻瘟霉活性,占分离菌株总数的32.2%。结论 5种药用植物中内生菌种类极其丰富,且有活性的菌株所占比例较高,HPLC检测有新的紫外吸收峰,值得对其次级代谢产物进一步研究。可见贵州药用植物具有丰富的活性内生菌资源,有巨大的开发研究价值。
Objective To isolate and screen endophytic bacteria with antibacterial activity and anti-tumor activity. Methods Five kinds of tissue materials including Rhubarb, Prickly Pear, Eucommia ulmoides, Radix Paeoniae Alba and Acanthopanax senticosus collected from Guizhou were isolated and sterilized by surface disinfection. The cells were separated by modified GYM medium and TWYE medium. Resistant Escherichia coli, drug-resistant Staphylococcus aureus, Micrococcus luteus, Streptococcus agalactiae and other test strains and Pyricularia oryzae strains were screened for the fermentation products of the isolated strains. The fermentation products of the active strains were further analyzed by HPLC. Results There were 180 endophytes isolated, of which 46 strains had antibacterial activity to varying degrees, accounting for 25.6% of the total isolates. 58 strains had different degree of anti-Pyricularia oryzae activity, accounting for 32.2% of the total isolates. Conclusion The five kinds of medicinal plants are extremely rich in endophyte species, and the proportion of active strains is high. New ultraviolet absorption peaks are detected by HPLC, and it is worth to further study their secondary metabolites. Visible Guizhou medicinal plants with rich active endophyte resources, a huge development and research value.
