建立了高效液相色谱同时测定苦楝子中的三种有效成分(香草酸、香兰素、阿魏酸)的方法。苦楝子样品用甲醇超声提取,提取液过滤并用甲醇稀释后分析。采用C18反相色谱柱(250×4.6nm,3.5μm),流动相为乙腈和0.1%醋酸水溶液,梯度洗脱。结果显示:三种有效成分的检出限(LOD)分别为2.5μg/m L、0.82μg/m L和1.3μg/m L,定量限(LOQ)分别为8.4μg/m L、2.7μg/m L和3.4μg/m L,加标回收率为94.9%~104.5%,相对标准偏差小于3.4%,苦楝子样品中的香草酸、香兰素、阿魏酸的平均含量分别是67.98 mg/kg、96.14 mg/kg、36.61 mg/kg(n=6),相对标准偏差分别为1.8%、1.1%和0.7%。该方法重复性好、简单、准确,可用于同时测定苦楝子中的香草酸、香兰素、阿魏酸的含量。
A method was established for simultaneous determination of three active ingredients (vanillic acid, vanillin, ferulic acid) in Melia azedarach by high performance liquid chromatography. Melia tomentosa samples with methanol ultrasonic extraction, the extract was filtered and diluted with methanol and analyzed. A C18 reversed-phase column (250 × 4.6 nm, 3.5 μm) was used with a mobile phase of acetonitrile and 0.1% aqueous acetic acid, gradient elution. The results showed that the limit of detection (LOD) of the three active ingredients were 2.5μg / m L, 0.82μg / m L and 1.3μg / m L, and the LOQ were 8.4μg / m L, 2.7μg / m L and 3.4 μg / m L, respectively. The recoveries were 94.9% -104.5% and the relative standard deviations were less than 3.4%. The average contents of vanillic acid, vanillin and ferulic acid in Melia tomentosa samples were 67.98 mg / kg, 96.14 mg / kg, 36.61 mg / kg respectively (n = 6). The relative standard deviations were 1.8%, 1.1% and 0.7% respectively. The method is reproducible, simple and accurate, and can be used for simultaneous determination of vanillic acid, vanillin and ferulic acid in Melia tomentosa.
