目的:建立小鼠组织中KS0604的定量分析方法。方法:采用组织消化结合基于离子交换和反相分配原理的两步固相萃取法,并采用无胶筛分毛细管电泳技术测定小鼠组织中的KS0604。结果:小鼠肾组织中KS0604在2.0~2000μg·g~(-1)范围内呈良好线性,最低定量限LOQ为2.0μg·g~(-1);其他组织中KS0604在1.3~128μg·g~(-1)范围内呈良好线性,最低定量限LOQ为1.3μg·g~(-1);r>0.999。加样回收率试验RSD<10.82%,精密度试验RSD<2.3%,稳定性试验结果表明KS0604在小鼠组织中的稳定性良好。结论:该方法可用于定量检测小鼠组织中的KS0604。
Objective: To establish a method for the quantitative analysis of KS0604 in mouse tissues. METHODS: Two-phase solid phase extraction (SPE) based on ion exchange and reversed phase partitioning was used in tissue digestion, and KS0604 in mouse tissues was determined by gel filtration-free capillary electrophoresis. Results: KS0604 showed a good linearity in the range of 2.0-2000 μg · g -1, and LOQ was 2.0 μg · g -1 in the lowest limit of quantitation (LOQ). In other tissues, KS0604 was in the range of 1.3-128 μg · g -1 ~ (-1), LOQ of the lowest limit of quantification was 1.3μg · g ~ (-1) and r> 0.999. RSD <10.82% for sample recovery test and RSD <2.3% for precision test. The results of stability test showed that the stability of KS0604 in mouse tissues was good. Conclusion: This method can be used to quantitatively detect KS0604 in mouse tissues.
