应用光谱方法研究了甘草酸二铵(DAG)与牛血清白蛋白之间的相互作用,探讨了甘草酸二铵对牛血清白蛋白构象的影响。结果表明,甘草酸二铵对牛血清白蛋白荧光具有较强的猝灭作用且为静态猝灭过程。根据Frster非辐射能量转移理论计算给体-受体间的距离(r=1.45 nm);得到结合常数(297K,K=1.14×105 L/mol;301K,K=7.99×104 L/mol)和结合位点数(297K,n=0.968;301K,n=0.932);研究了常见金属离子和甘草次酸(GA)对该体系相互作用的影响,结果表明Co2+对DAG与BSA的结合起到明显的正协同作用,Mn2+、Zn2+和GA对DAG与BSA的结合有明显竞争作用。
The interaction between diammonium glycyrrhizinate (DAG) and bovine serum albumin (BSA) was studied by spectroscopic method and the effect of diammonium glycyrrhizinate on the conformation of bovine serum albumin was discussed. The results show that diammonium glycyrrhizinate has a strong quenching effect on bovine serum albumin fluorescence and is a static quenching process. The donor-acceptor distance (r = 1.45 nm) was calculated according to the Frster non-radiative energy transfer theory; the binding constant (297K, K = 1.14 × 105 L / mol; 301K, K = 7.99 × 104 L / mol ) And the number of binding sites (297K, n = 0.968; 301K, n = 0.932). The effects of common metal ions and glycyrrhetinic acid (GA) on the interaction were studied. The results showed that the binding of DAG to BSA Significant positive synergistic effect, Mn2 +, Zn2 + and GA on the binding of DAG and BSA obvious competition.
