以优良植物生防菌解淀粉芽孢杆菌L-S60为研究对象,通过单因素试验研究酸碱、盐浓度、温度对该菌株生长的影响;检测解淀粉芽孢杆菌L-S60的产酶和抑菌特性;并研究优化解淀粉芽孢杆菌L-S60的固态发酵工艺。结果表明:解淀粉芽孢杆菌L-S60具有较宽的酸碱耐受性,盐的质量分数>10%时菌落数会显著下降,80℃以下菌落数无明显变化。解淀粉芽孢杆菌L-S60能产生蛋白酶、纤维素酶和淀粉酶,且对多种病原真菌有抑制作用。L-S60的最佳固体发酵条件为:温度35℃、接种量30%、料水质量比1∶0.45、装瓶量40%和发酵时间42h;最佳固体培养基组成为,m(花生饼粉)∶m(麦麸)∶m(棉粕)=53∶35∶12,外源物质蛋白胨、葡萄糖和MgSO4·7H2O添加量分别为3.00、2.00和0.20g/kg,此时,解淀粉芽孢杆菌L-S60菌落数达到2.42×1010 CFU/g。解淀粉芽孢杆菌L-S60具有很好的开发利用价值。
To study the effect of acid-base, salt concentration and temperature on the growth of Bacillus amyloliquefaciens L-S60 by single-factor experiment, the enzyme production and antibacterial activity of Bacillus amyloliquefaciens L-S60 Characteristics; and optimize the solid Bacillus amyloliquefaciens L-S60 fermentation process. The results showed that Bacillus amyloliquefaciens L-S60 had wide acid-base tolerance. The number of colonies decreased significantly when the salt concentration was> 10%, and the number of colonies under 80 ℃ did not change significantly. Bacillus amyloliquefaciens L-S60 can produce proteases, cellulases and amylases, and has an inhibitory effect on a variety of pathogenic fungi. The optimal solid fermentation conditions of L-S60 were as follows: temperature 35 ℃, inoculation amount 30%, mass ratio of material to water 1: 0.45, bottling 40% and fermentation time 42h. The best solid medium consisted of m Powder): m (wheat bran): m (cottonseed meal) = 53:35:12, exogenous peptone, glucose and MgSO4 · 7H2O were 3.00,2.00 and 0.20g / kg, respectively. At this time, Bacillus L-S60 colonies reached 2.42 × 1010 CFU / g. Bacillus amyloliquefaciens L-S60 has good development and utilization value.
