目的:探讨补阳还五汤对阿尔茨海默病小鼠学习记忆能力及海马β淀粉样前体蛋白(APP)的影响。方法:APP/PS1双转基因小鼠随机分成对照组、模型组、治疗组(吡拉西坦0.62 g·kg-1)、补阳还五汤高剂量组(37.06 g·kg-1)、补阳还五汤中剂量组(18.53 g·kg-1)、补阳还五汤低剂量组(9.26 g·kg-1)。给予补阳还五汤ig,连续治疗28 d后取材。采用Morris水迷宫测试行为学指标,采用双抗体夹心法测定海马组织APP蛋白水平。结果:与对照组比较,模型组小鼠逃避潜伏期延长(P<0.05),游泳距离增加(P<0.05),APP水平增加(P<0.05);与模型组比较,补阳还五汤能改善模型小鼠的学习和记忆(P<0.05),并且降低海马APP水平(P<0.05)。结论:补阳还五汤能改善模型小鼠的学习记忆功能,能够通过抑制APP蛋白在阿尔茨海默病治疗中发挥重要的作用。
Objective: To investigate the effect of Buyang Huanwu Decoction on learning and memory abilities and hippocampal β-amyloid precursor protein (APP) in mice with Alzheimer’s disease. Methods: APP / PS1 double transgenic mice were randomly divided into control group, model group, treatment group (piracetam 0.62 g · kg -1), BUYH high dose group (37.06 g · kg -1) Yang-huanwu decoction middle-dose group (18.53 g · kg-1), Buyang Huanwu decoction low-dose group (9.26 g · kg-1). Give Yang also replenishing the soup ig, continuous treatment of 28 d drawn. Morris water maze test behavioral indicators, using double antibody sandwich method to measure the level of APP protein in the hippocampus. Results: Compared with the control group, the mice in the model group had longer escape latency (P <0.05), longer swimming distance (P <0.05), and higher level of APP (P <0.05). Compared with the model group, BYHWD (P <0.05), and decreased the level of APP in hippocampus (P <0.05). Conclusion: Buyang Huanwu decoction can improve learning and memory in model mice and play an important role in the treatment of Alzheimer’s disease by inhibiting APP protein.
