探讨正常 SD大鼠血清及组织内源性哇巴因 ( EO)的含量 ,为 EO生理及病理作用的进一步研究提供理论及实验依据。方法 制备高度特异性的哇巴因抗体 ,应用酶联免疫吸附试验 ( ELISA)的方法 ,对正常 SD大鼠血清及组织标本测定其 EO含量。结果 血清、肝脏、肾脏、肾上腺、垂体及下丘脑内 EO含量分别为 ( 1 .1 2± 0 .1 7) μg/ L、( 1 .79± 0 .44) μg/ kg组织、( 1 .0 8±0 .63)μg/ kg组织、( 1 .73± 0 .81 )μg/ kg组织、( 2 7.54± 6.91 )μg/ kg组织、( 1 .83± 0 .54)μg/ kg组织、( 1 0 .1 0± 3.0 8) μg/ kg组织。肾上腺内 EO含量明显高于其它组织及血清 EO含量。结论 确定了正常 SD大鼠血清及多种组织内源性哇巴因的含量 ,提示肾上腺有可能是 EO的主要来源
To investigate the content of endogenous ouabain (EO) in serum and tissue of normal SD rats, and to provide theoretical and experimental evidences for the further study on physiological and pathological effects of EO. Methods The highly specific ouabain antibody was prepared and the serum and tissue samples of normal SD rats were assayed for EO content by enzyme linked immunosorbent assay (ELISA). Results The contents of EO in serum, liver, kidney, adrenal gland, pituitary and hypothalamus were (1.12 ± 0.17) μg / L and (1.79 ± 0.44) μg / kg, respectively 0 8 ± 0. 63) μg / kg tissue, (1.73 ± 0.81) μg / kg tissue, (2 7.54 ± 6.91) μg / kg tissue, and (1.83 ± 0.54) μg / kg tissue , (100.0 ± 3.08) μg / kg tissue. Adrenal EO content was significantly higher than other tissues and serum EO content. Conclusion The levels of endogenous ouabain in serum and various tissues of SD rats were determined, suggesting that adrenal gland may be the main source of EO
