目的建立特异、快速、准确的SARS冠状病毒检测方法 ,并探讨其临床应用价值。方法根据WHO公布的SARS冠状病毒的基因序列 ,采用套式RT -PCR方法对 12 7份SARS患者不同病程的血清标本中SARS病毒核酸进行扩增 ,用敏感度 ,特异度 ,阳性预测值 (PPV) ,阴性预测值 (NPV) ,准确度等对该方法诊断SARS的效果进行评价。结果病程在 10d以内的 32份血清标本中 ,13份阳性 ,敏感度 4 0 .6 % ,特异度 10 0 .0 % ,阳性预测值 10 0 .0 % ,阴性预测值 6 6 .1% ,准确度 72 .5 %。结论本研究建立的套式RT -PCR方法可作为发病早期SARS病人的辅助诊断。
Objective To establish a specific, rapid and accurate SARS-CoV detection method and to explore its clinical value. Methods According to the sequence of WHO published SARS coronavirus, SARS virus nucleic acid was amplified by using the method of nested RT-PCR in sera of 12 7 SARS patients with different course of disease. The sensitivity, specificity, positive predictive value (PPV) ), Negative predictive value (NPV), accuracy and so on to evaluate the effect of this method in the diagnosis of SARS. Results Among the 32 serum samples with the course of 10 days, 13 were positive, the sensitivity was 40.6%, the specificity was 100.0%, the positive predictive value was 100.0%, the negative predictive value was 6.61% Accuracy of 72.5%. Conclusion The nested RT-PCR method established in this study can be used as an auxiliary diagnosis of SARS patients with early onset.
