目的观察淫羊藿苷对维生素D缺乏大鼠血清25(OH)D_3、1,25(OH)D_3、皮质醇及肾上腺皮质CYP17A1mRNA表达的影响。方法选用60只雄性SD大鼠,随机分为正常组,模型组[生理盐水4.0ml/(kg·d)灌胃],淫羊藿苷高剂量组[120mg/(kg·d)灌胃],淫羊藿苷中剂量组[60mg/(kg·d)灌胃],淫羊藿苷低剂量组[30mg/(kg·d)灌胃],维生素D组[30mg/(kg·d)灌胃]。造模1月后,药物干预2月,酶联免疫法(ELISA法)检测血清25(OH)D_3、1,25(OH)D_3、皮质醇;Real-Time PCR法检测肾上腺皮质CYP17A1mRNA表达。结果与正常组比较,模型组血清25(OH)D_3、1,25(OH)D3含量下降(P<0.05),皮质醇含量下降(P<0.05);淫羊藿苷干预后血清25(OH)D_3、1,25(OH)D3、皮质醇含量均上升(P<0.05)。Real-Time PCR显示,模型组肾上腺皮质CYP17A1mRNA表达量下调(P<0.05),淫羊藿苷干预后表达量均上调(P<0.05)。结论淫羊藿苷可能改善维生素D缺乏大鼠皮质醇的分泌。
Objective To observe the effect of icariin on serum 25 (OH) D_3, 1, 25 (OH) D_3, cortisol and adrenocortical CYP17A1mRNA in vitamin D deficiency rats. Methods Sixty male Sprague-Dawley rats were randomly divided into normal group, model group [normal saline 4.0ml / (kg · d)), high dose icariin group [120mg / (kg · d) , Icariin medium dose group [60mg / (kg · d)), icariin low dose group [30mg / (kg · d)) and vitamin D group [30mg / (kg · d) Gavage]. After modeling for one month, serum 25 (OH) D_3, 1, 25 (OH) D_3 and cortisol were detected by enzyme-linked immunosorbent assay (ELISA) and CYP17A1 mRNA expression in adrenal cortex were detected by Real-Time PCR. Results Compared with the normal group, the content of serum 25 (OH) D 3, 1, 25 (OH) D3 in the model group decreased (P <0.05) and the content of cortisol decreased (P < ) D_3,1,25 (OH) D3, cortisol content increased (P <0.05). Real-Time PCR showed that the expression of CYP17A1 mRNA in adrenal cortex of model group was down-regulated (P <0.05), and the expression of icariin was up-regulated after intervention (P <0.05). Conclusion Icariin may improve the secretion of cortisol in vitamin D-deficient rats.
