目的:建立HPLC法同时测定中药小蓟中绿原酸、咖啡酸、黄芩苷、芦丁、蒙花苷、刺槐素6种活性成分的含量。方法:Diamonsil C18(4.6 mm×200 mm,5μm)色谱柱,流动相为甲醇(A)-0.3%磷酸水溶液(B),梯度洗脱,流速:1 m L/min,检测波长:327 nm,柱温:30℃。结果:绿原酸、咖啡酸、黄芩苷、芦丁、蒙花苷和刺槐素的质量浓度分别在0.0520~2.0500μg(r=0.9995)、0.0040~0.1700μg(r=0.9997)、0.0180~0.7320μg(r=0.9998)、0.0300~1.2120μg(r=0.9997)、0.0500~2.0200μg(r=0.9999)、0.0100~0.3820μg(r=0.9998)内与峰面积呈良好的线性关系。绿原酸、咖啡酸、黄芩苷、芦丁、蒙花苷和刺槐素的平均加样回收率分别为97.56%、98.59%、97.62%、99.97%、99.47%和99.04%,RSD%(n=6)分别为1.17%、1.60%、1.41%、1.66%、0.93%和1.45%。结论:该方法简便、准确、重复性好,可用于小蓟药材多成分的质量控制研究。
OBJECTIVE: To establish an HPLC method for the simultaneous determination of six active constituents of chlorogenic acid, caffeic acid, baicalin, rutin, monolin glycoside and acacia in Chinese thistle. METHODS: Diamonsil C18 (4.6 mm × 200 mm, 5 μm) was used as the mobile phase. The mobile phase consisted of methanol (A) -0.3% phosphoric acid solution (B) with gradient elution at a flow rate of 1 mL / Column temperature: 30 ℃. Results: The concentrations of chlorogenic acid, caffeic acid, baicalin, rutin, mandarin and acacia were 0.0520 ~ 2.0500μg (r = 0.9995), 0.0040 ~ 0.1700μg (r = 0.9997), 0.0180 ~ 0.7320μg (r = 0.9998), 0.0300 ~ 1.2120μg (r = 0.9997), 0.0500 ~ 2.0200μg (r = 0.9999) and 0.0100 ~ 0.3820μg (r = 0.9998) The average recoveries of chlorogenic acid, caffeic acid, baicalin, rutin, magenta and acacia were 97.56%, 98.59%, 97.62%, 99.97%, 99.47% and 99.04%, respectively. The RSD% 6) were 1.17%, 1.60%, 1.41%, 1.66%, 0.93% and 1.45% respectively. Conclusion: The method is simple, accurate and reproducible. It can be used to study the quality control of multi-component medicinal materials of Artichoke.
