目的 研究白细胞介素17A(IL-17A)对右侧星状神经节(RSG)功能和神经活性的影响.方法 16只健康成年杂种犬静脉麻醉后随机分为实验组(n=8)和对照组(n=8).分离RSG,在实验组中,向RSG中注射0.1ml IL-17A (25 μg/ml),对照组注射等量的生理盐水.在基础状态和注射后30 min后分别测定高频电刺激(HFS:频率20 Hz,脉宽0.1 ms)RSG时的心率变化以及RSG神经活性.同时实验结束后留取RSG组织用于检测早期原癌基因(c-fos)和神经生长因子(NGF)表达水平.结果 实验组干预后心率加快作用明显增强(40 V:57%±9%vs 40%±8%,P<0.05),RSG神经活性显著增强[频率:(148±22)次/分vs (48+ 10)次/分;幅度:(0.09±0.01)mV vs(0.04±0.01)mV,P均<0.05],同时RSG组织中c-fos和NGF表达水平明显升高(c-fos:0.58±0.08 vs 0.55±0.09;NGF:0.16±0.04 vs 0.11±0.03,P均<0.05);而对照组以上指标无明显改变.结论 IL-17A可增强RSG功能和神经活性,增加神经相关蛋白表达水平.“,”Objective To investigate the effect of proinflammatory cytokine interleukin-17A(IL-17A) on the neural function and activity of right stellate ganglion in normal canine.Methods Sixteen healthy canines were randomly divided into experimental group (n =8) and control group (n =8) after intravenous anesthesia.In experimen tal group,the right stellate ganglion was separated and 0.1 ml IL-17A (25 μg/ml) was injected into the ganglion,while 0.1 ml saline was injected in control group.The change of heart rate in high frequency electrical stimulation (HFS:frequency 20 Hz,pulse width 0.1 ms) on RSG and RSG nerve activity were determined at baseline and 30 minute after injection.The RSG tissue was resected for detecting the protein expression levels of c fos and NGF.Results In experimental group,the change of heart rate (40 V:57%±9% vs 40%±8%,P<0.05),the frequency [(148±22) impulses/min vs (48±10) impluses/min,P<0.05] and amplitude [(0.09±0.01)mV vs (0.04±0.91) mY,P<0.05] of RSG neural activity were markly increased after IL 17A injection.Additionally,IL-17A injection upregulated the protein expression level of c-ffos and NGF when compared with the control group (c-fos:0.58 ± 0.08 vs 0.55±0.09;NGF:0.16±0.04 vs 0.11±0.03,all P<0.05).Conclusion IL 17A could enhance the function and activity of RSG and upregulate the protein expression levels of c-fos and NGF.
