目的探讨急性缺血性脑血管病血小板肌球蛋白轻链激酶(MLCK)和Ca2+,Mg2+-ATP酶活性的变化及钙拮抗剂对Ca2+,Mg2+-ATP酶活性的影响。方法用32P同位素掺入法和比色法分别测定58例脑中风病人,包括22例短暂性脑缺血发作(TIA)患者和36例脑梗塞患者以及35名健康对照者血小板MLCK和Ca2+,Mg2+-ATP酶活性。结果TIA组和脑梗塞组MLCK活性与对照组相比均有明显增加(P<0.01),而Ca2+,Mg2+-ATP酶活性均低于对照组(P<0.05,P<0.01)。在TIA组和脑梗塞组之间,这两种酶活性无明显差异(P>0.05)。用钙拮抗剂nimodipine治疗后,TIA组和脑梗塞组患者Ca2+,Mg2+-ATP酶活性均有部分恢复(P<0.05)。结论血小板MLCK和Ca2+,Mg2+-ATP酶活性与急性缺血性脑血管病的发生有密切关系,钙拮抗剂可部分改善血小板Ca+2,Mg2+-ATP酶活性状态。
Objective To investigate the changes of platelet myosin light chain kinase (MLCK) and Ca2 +, Mg2 + -ATPase activity in acute ischemic cerebrovascular disease and the effects of calcium antagonists on Ca2 +, Mg2 + -ATPase activity. Methods Fifty-eight stroke patients, including 22 transient ischemic attacks (TIA), 36 cerebral infarction patients and 35 healthy controls, were assayed for platelet MLCK and Ca2 +, Mg2 + -ATPase activity. Results The activity of MLCK in TIA group and cerebral infarction group were significantly higher than those in control group (P <0.01), while the activity of Ca2 + and Mg2 + -ATP was lower than that in control group (P <0.05, P <0.01). 01). There was no significant difference in activity between TIA group and cerebral infarction group (P> 0.05). After treatment with calcium antagonist nimodipine, the activity of Ca2 +, Mg2 + -ATP in TIA group and cerebral infarction group were partially recovered (P <0.05). Conclusion Platelet MLCK and Ca2 +, Mg2 + -ATPase activity are closely related to the occurrence of acute ischemic cerebrovascular disease. Calcium antagonists can partially improve the activity of platelet Ca 2+, Mg 2+ -ATPase.
