目的大量研究表明高血压病是一种炎症性疾病,本课题组前期研究从淋巴细胞钾通道水平初步揭示高血压可能是一种炎症性疾病,但具体机制不清。故本研究通过研究新疆哈萨克族单纯高血压病患者外周血淋巴细胞CaN/NFAT信号通路发生的变化,拟揭示钾通道参与高血压病发生发展的具体机制。方法选取2014年2月—2014年11月首诊在新疆医科大学第一附属医院心脏中心的单纯高血压病未使用药物降压治疗的新疆哈萨克族患者。采集静脉血,采用ELISA技术检测hsCRP,根据hsCRP水平将研究对象分为3组:高hsCRP高血压组、低hsCRP高血压组和正常血压对照组,每组各50例。采集外周血分离淋巴细胞。采用Real-time PCR定量技术、Western blot技术检测Kv1.3、TNF-α基因和蛋白表达水平,CaN、NFAT活性。结果高hsCRP高血压组和低hsCRP高血压组Kv1.3、TNF-αmRNA、蛋白表达水平及其CaN/NFAT活性高于正常血压对照组(P<0.05),差异有统计学意义。高hsCRP高血压组Kv1.3、TNF-αmRNA、蛋白表达水平及其CaN/NFAT活性表达水平高于低hsCRP高血压组,差异有统计学意义(P<0.05)。结论新疆哈萨克族单纯高血压病患者CaN/NFAT信号通路活性增加,钾通道可能通过CaN/NFAT信号通路参与高血压病的发生发展。
PURPOSE A large number of studies have shown that hypertension is an inflammatory disease. Our preliminary study revealed that hypertension may be an inflammatory disease from the level of lymphocytes potassium channel, but the specific mechanism is unclear. Therefore, this study aims to reveal the specific mechanism by which potassium channel participates in the development of hypertension by studying the changes of CaN / NFAT signaling pathway in peripheral blood lymphocytes in patients with essential hypertension in Xinjiang Kazakhs. Methods From February 2014 to November 2014, Kazak people from Xinjiang who were treated with antihypertensive drugs without hypertension were enrolled in the study of hypertension in the heart center of the First Affiliated Hospital of Xinjiang Medical University. The venous blood was collected and the hsCRP was detected by ELISA. According to the level of hsCRP, the subjects were divided into three groups: high hsCRP high blood pressure group, low hsCRP high blood pressure group and normal blood pressure control group with 50 cases in each group. Collect peripheral blood to separate lymphocytes. The expression of Kv1.3, TNF-α gene and protein, CaN, NFAT activity were detected by Real-time PCR and Western blot. Results The expressions of Kv1.3, TNF-αmRNA, protein and CaN / NFAT in high hsCRP group and low hsCRP group were higher than those in normotensive group (P <0.05). The difference was statistically significant. The expression levels of Kv1.3, TNF-αmRNA, protein and CaN / NFAT in high-hsCRP group were significantly higher than those in low-hsCRP group (P <0.05). Conclusion The activity of CaN / NFAT signaling pathway in patients with essential hypertension in Kazak ethnic group in Xinjiang is increased. Potassium channel may participate in the development of hypertension by CaN / NFAT signaling pathway.
