本文报道了甲基亚硝基脲(methylnitrosourea,MNU)诱发RF/J系小鼠T细胞淋巴瘤在裸小鼠(nudemice)体内连续传代的染色体变化。我们选用4个T细胞淋巴瘤接种6只裸小鼠而建立6个细胞系,每系连续传代5~7次。其核型分析结果九有11种异常染色体的发生率比预计的要高,按递减的顺序分别为10、X、3、12、6、1、4、19、15、18、14。染色体异常发生率随传代培养逐渐增加。如果MNU只是选择性地作用于某些特定的基因位点,这对解释其致癌机理有重要意义。
In this paper, we report the chromosomal changes of RFL mice induced by methylnitrosourea (MNU) in nude mice. We selected 6 T-cell lymphoma inoculated 6 nude mice and the establishment of 6 cell lines, each line of continuous passage 5 to 7 times. The karyotype analysis showed that the incidence of 11 abnormal chromosomes was higher than expected, decreasing by 10, X, 3, 12, 6, 1, 4, 19, 15, 18 and 14, respectively. The incidence of chromosomal abnormalities gradually increased with subculture. If MNU selectively acts on certain specific loci, it is important to explain its carcinogenic mechanism.
