目的观察生骨胶囊对兔骨折愈合过程中转化生长因子-β1(TGF-β1)的影响,以进一步探讨其促进骨折愈合的机制。方法将60只健康雄性家兔随机分为治疗组和对照组各30只,在兔桡骨处用手术方法制备骨折模型,造模后即日治疗组予生骨胶囊150mg/(kg.d),对照组予骨折挫伤胶囊150mg/(kg.d)。于骨折后3d和1、2、3、4、6周每组分别抽取5只兔处死,取标本,采用免疫组化法,并利用图像分析的方法测定TGF-β1表达强度。结果骨折早期TGF-β1染色强度均值低,以后逐渐升高,成骨活跃时最高,随着修复逐渐完成,TGF-β1染色强度均值逐渐降低。骨折后3d,相当于骨折损伤反应期,对照组TGF-β1染色强度均值较治疗组高(P<0.05);治疗组在骨折后2周出现染色高峰,较对照组提前1周,比较差异有统计学意义(P<0.01)。结论生骨胶囊可以调节骨折愈合过程中TGF-β1的合成和分泌,从而促进骨折愈合,TGF-β1可能是生骨胶囊对骨折愈合过程中的一个作用环节。
Objective To observe the effect of Shenggu capsule on the expression of transforming growth factor-β1 (TGF-β1) during fracture healing in rabbits to further explore its mechanism of promoting fracture healing. Methods Sixty healthy male rabbits were randomly divided into treatment group and control group, with 30 rabbits in each group. The fracture models were prepared by surgery in rabbits’ radius and treated with Shenggu capsule 150mg / (kg · d) Group was given a contusion and contusion capsule 150mg / (kg.d). Five rabbits in each group were sacrificed on the 3rd, 3rd, 4th, 4th, 6th week after the fracture, and the specimens were taken for immunohistochemistry. The intensity of TGF-β1 expression was determined by image analysis. Results The mean value of TGF-β1 staining intensity in the early stage of fractures was low, then increased gradually, and reached the peak at the time of osteogenesis. As the repair was completed, the mean value of TGF-β1 staining intensity gradually decreased. Compared with the treatment group, the mean value of TGF-β1 staining intensity in the control group was higher than that in the treatment group after 3d fracture fracture (P <0.05), and the staining peak appeared two weeks after the fracture in the treatment group, one week earlier than the control group Statistical significance (P <0.01). Conclusion Shengjian Capsule can regulate the synthesis and secretion of TGF-β1 in the process of fracture healing, thereby promoting fracture healing. TGF-β1 may be an important part in the process of fracture healing.
