目的观察高脂诱导的胰岛素抵抗对大鼠勃起功能的影响。方法以有正常勃起功能的雄性SD大鼠为研究对象,分为正常对照组和高脂组,高脂喂养制备胰岛素抵抗模型。成模后测定各组大鼠隔夜取血测空腹血糖(fasting blood glucose,FBG)、胰岛素(fasting plasma insulin,FINS)、胆固醇(total cholesterol,TC)、低密度脂蛋白胆固醇(low density lipoprotein cholesterol,LDL-C)、甘油三酯(triglyceride,TG)、游离脂肪酸(free fatty acids,FFA)、丙二醛(malondialdehyde,MDA)、超氧化物歧化酶(superoxide dismutase,SOD)、一氧化氮(nitric oxide,NO);并行阿朴吗啡(apomorphine,APO)试验测定大鼠勃起功能;取大鼠阴茎海绵体平滑肌匀浆,测定MDA、SOD、NO水平。结果 1与对照组比较高脂组大鼠体质量、FINS(4.3±0.7)μU/ml、胰岛素抵抗指数(insulin resistance index,Homa-IR)均升高(P<0.05),造模成功;血FFAs、TG、LDL-C、MDA均明显升高(P<0.05),SOD、NO降低(P<0.05);阴茎海绵体平滑肌MDA、SOD、NO与血清中的变化一致。2高脂组中有13只大鼠出现勃起障碍,勃起率为56.67%,低于正常对照组的100%(P<0.05)。结论高脂喂养可致大鼠出现勃起功能障碍,其机制可能与胰岛素抵抗所致氧化应激损伤有关。
Objective To observe the effect of high fat-induced insulin resistance on erectile function in rats. Methods Male Sprague-Dawley rats with normal erection function were divided into normal control group and high fat diet group, and insulin resistance model was established by high fat diet. The fasting blood glucose (FBG), fasting plasma insulin (FINS), total cholesterol (TC) and low density lipoprotein cholesterol LDL-C, triglyceride (TG), free fatty acids (FFA), malondialdehyde (MDA), superoxide dismutase (SOD), nitric oxide (NO). The apomorphine (APO) test was used to measure the erectile function in rats. The smooth muscle of rat corpus cavernosum was taken for determination of MDA, SOD and NO. Results Compared with the control group, the body weight, FINS (4.3 ± 0.7) μU / ml and insulin resistance index (Homa-IR) FFAs, TG, LDL-C and MDA were significantly increased (P <0.05), while SOD and NO were decreased (P <0.05). The contents of MDA, SOD and NO in the corpus cavernosum were the same as those in serum. Eighteen rats with hyperlipidemia had erectile dysfunction with an erection rate of 56.67%, which was lower than that of the normal control group (P <0.05). Conclusion High fat diet can cause erectile dysfunction in rats. The mechanism may be related to the oxidative stress injury induced by insulin resistance.
