目的:制备高纯度的兔抗SARS冠状病毒的IgG。方法:采用辛酸-硫酸铵沉淀进行初纯,以及DEAE离子交换层析精纯IgG,利用SDS-PAGE对纯化后的IgG纯度进行测定,利用间接ELISA、病毒中和滴定对IgG进行效价测定。结果:纯化后的IgG相对纯度达到90%以上,ELISA检测效价为1∶4 800,病毒中和效价为1∶640。结论:利用上述纯化工艺能获得高纯度的兔抗SARS冠状病毒IgG,同时抗体活性损失较小。
Objective: To prepare high purity rabbit anti-SARS coronavirus IgG. Methods: Purification was performed by using octanoic acid-ammonium sulfate precipitation, and purified by DEAE ion exchange chromatography. The purity of purified IgG was determined by SDS-PAGE. The titer of IgG was determined by indirect ELISA and virus neutralization titration. Results: The purity of the purified IgG was over 90%. The ELISA titer was 1: 800 and the virus titer was 1: 640. Conclusion: The rabbit anti-SARS coronavirus IgG with high purity can be obtained by the above-mentioned purification process, and the antibody activity loss is small at the same time.
