目的:研究尿激酶( U K) 或蚓激酶( E K) 与氟罗沙星( F L R X) 联合应用对细菌生物被膜的影响。方法:最低抑菌浓度测定采用标准微量稀释法;以银染法快速鉴定细菌生物被膜( B B F) ;扫描电镜( S E M) 观察 B B F 形态;用甲基四氮唑蓝( M T T) 比色法测定 B B F 中活菌数。结果:经 F L R X 处理 B B F 的 S E M 图像示细菌形态仍保持短杆状,但细菌数量及菌体间纤维样黏液物质显著减少;经 U K 或 E K 与 F L R X 合用处理后的 B B F 图像显示细菌破碎、形态不一,菌数和菌体间粘液丝均比单用 F L R X 时明显减少。对活菌数影响与电镜图像结果一致。结论: U K 或 E K 与 F L R X 合用对铜绿假单胞菌 B B F 的抑制具有明显的协同效应。
OBJECTIVE: To study the effects of urokinase (U K) or lumbrokinase (E K) and fleroxacin (F L R X) on bacterial biofilm. Methods: The minimum inhibitory concentration (MIC) was determined by standard microdilution method; the bacterial biofilm (B B F) was identified by silver staining; the morphology of B B F was observed by scanning electron microscopy (SEM) T T) colorimetric determination of viable bacteria in B B F. Results: The SEM images of B B F treated with F L R X showed that the morphology of the bacteria remained short rod-shaped, but the number of bacteria and interstitial fibrillar mucus were significantly reduced. After U K or E K and F L R X B B F images after combined treatment showed that the bacteria were broken, the morphology was different, and the number of bacteria and the mucilage filaments in the cells were significantly reduced compared with that of the FRL alone. The number of viable cells and electron microscopy image results. Conclusion: Combination of U K or E K with F L R X has obvious synergistic effect on the inhibition of B F of Pseudomonas aeruginosa.
