目的至今,尚未见到采用免疫组化技术研究骨髓辐射损伤后干细胞因子受体(C-kit)表达及其在造血重建中的意义。方法经60Coγ射线全身照射LACA小鼠,于照射后4周内分批活杀动物,采用光镜、电镜及LSAB免疫组化方法,研究了辐射诱导的造血细胞凋亡及C-kit的表达及意义。结果全身照射LACA小鼠时,5.5Gy为其亚致死剂量,此剂量照射小鼠可导致外周血白细胞及骨髓有核细胞数减少;骨髓明显损伤及损伤后重建现象;在照后1天内可使骨髓造血细胞发生凋亡;5.5Gy照射后6小时,C-kit蛋白于造血细胞浆内明显增多,呈强阳性;照后10天,骨髓造血细胞增生、恢复,C-kit蛋白于造血细胞膜表达明显增多,呈强阳性。结论C-kit表达与造血功能重建有关,在早期造血功能重建中起重要调节作用。

On the 10th day after irradiation, bone marrow hematopoietic cells proliferated and recovered. C-kit protein was found to hematopoietic Cell membrane expression was significantly increased, was strongly positive. Conclusions C-kit expression is related to hematopoietic reconstitution and plays an important role in early hematopoietic reconstitution.
