[目的]探讨马钱子碱对乳腺癌骨转移相关因子的影响。[方法]建立裸鼠乳腺癌骨转移的模型,并用马钱子碱干预后用逆转录聚合酶链反应法(RT-PCR)检测各组肿瘤组织中骨转移相关因子MMP-2、CXCR4、RANKL、OPCmRNA表达量。[结果]马钱子碱经腹腔给药的半数致死量(Medial lethal dose,JLD_(50))为69mg/kg。根据LD_(50)的1/10、1/20和1/40的量,设三个剂量组:高剂量组6.90mg/kgl、中剂量组3.45mg/kg和低剂量组1.73mg/kg,同时与空白对照组比较。马钱子碱各剂量组MMP-2,CXCR4,RANKL mRNA的表达量随着马钱子碱剂量的增加逐渐下降(P<0.05),而OPG mRNA的表达量则随着马钱子碱剂量的增加逐渐增加(P<0.05)。[结论]马钱子碱可以抑制乳腺癌骨转移,主要以调节MMP-2、CXCR4、RANKL、OPG等骨转移相关因子的表达为主要手段。
[Objective] To investigate the effect of brucine on bone metastasis related factors in breast cancer. [Methods] The model of bone metastasis of breast cancer was established in nude mice. The expression of MMP-2, CXCR4, RANKL, MMP-2 and MMP-9 were detected by reverse transcription polymerase chain reaction , OPC mRNA expression level. [Result] The median lethal dose (JLD 50) of strychnine administered intraperitoneally was 69 mg / kg. According to the 1/10, 1/20 and 1/40 LD_ (50), three dose groups were set up: 6.90mg / kg for the high dose group, 3.45mg / kg for the middle dose group and 1.73mg / kg for the low dose group, At the same time compared with the blank control group. The expression of MMP-2, CXCR4 and RANKL mRNA in each dose of strychnine decreased with the increase of brucine dose (P <0.05), while the expression of OPG mRNA increased with the dose of brucine Increase gradually (P <0.05). [Conclusion] Brucine can inhibit the bone metastasis of breast cancer, and mainly regulate the expression of MMP-2, CXCR4, RANKL, OPG and other bone metastasis-related factors.
