目的研究葛根-黄连药对提取物对α-葡萄糖苷酶活性及大鼠餐后高血糖的抑制作用。方法提取大鼠小肠α-葡萄糖苷酶,以蔗糖为底物,用葡萄糖氧化酶法研究葛根-黄连药对、葛根和黄连单味药材提取物对α-葡萄糖苷酶的抑制活性。以蔗糖诱导的餐后高血糖大鼠为模型,测定灌胃给予葛根-黄连药对提取物后的血糖值,研究葛根-黄连药对对大鼠餐后高血糖的抑制作用。结果葛根-黄连药对、葛根和黄连提取物对α-葡萄糖苷酶的抑制作用呈剂量依赖性,IC50分别为2.439、3.700和5.730 g·L~(-1),葛根-黄连药对提取物对该酶的抑制作用最强;高剂量组的葛根-黄连药对提取物(400 mg·kg~(-1))可显著降低大鼠进食蔗糖后的血糖值。结论葛根-黄连药对提取物对α葡萄糖苷酶的抑制作用显著强于单味药材提取物,并可显著缓解大鼠餐后血糖升高。
Objective To study the inhibitory effect of Pueraria - Coptis Chinensis Franch on the activity of α-glucosidase and postprandial hyperglycemia in rats. Methods The intestinal α-glucosidase was extracted from rat intestine. The glucose-oxidase method was used to study the inhibitory activity of the Pueraria - Coptis Chinensis herb extract and Puerariae and Coptis Chinensis officinalis extracts on the α-glucosidase activity. The model of postprandial hyperglycemia induced by sucrose was used to determine the blood glucose level of Puerarin-Coptidis Rhizoma Coptidis-Coptis-Rhizoma Coptidis after oral administration. The inhibitory effect of Pueraria-Coptis-Rhizoma Coptis on postprandial hyperglycemia was studied. Results Puerarin - Coptis Chinensis, Radix Puerariae and Coptis Extract had a dose - dependent inhibitory effect on α - glucosidase with IC50 of 2.439, 3.700 and 5.730 g · L -1, The inhibitory effect of this enzyme was the strongest; Pueraria - Coptis extract (400 mg · kg -1) in high dose group could significantly reduce the blood glucose level after sucrose intake. Conclusion Puerarin - Coptis against the α - glucosidase extract was significantly stronger than the single herb extract, and significantly alleviate postprandial blood glucose levels in rats.
