目的 探讨虎杖苷对心肌细胞收缩性的直接影响及其作用机制。方法 原代培养幼鼠心肌细胞 ,利用计算机图象分析系统测定虎杖苷处理后心肌细胞收缩频率和收缩幅度的变化。结果 当加入 0 1mmol·L-1虎杖苷到培养细胞中时 ,心肌细胞收缩频率显著加快 ,收缩幅度增强 ;当加入洋地黄类药物西地兰时 ,心肌细胞收缩幅度增强 ,搏动频率也加快。而加入D Hanks液的对照组心肌细胞搏动情况在 30min内未发生变化。给予虎杖苷后 ,心肌细胞内钙荧光强度呈现进行性上升趋势 ,上升到最初的 2 2 4 0 %± 2 4 33%。结论 虎杖苷可能通过促使心肌细胞内钙离子浓度升高而直接增强心肌细胞的收缩性
Objective To investigate the direct effect of polydatin on myocardial contractility and its mechanism of action. Methods Cardiomyocytes from young rats were cultured for the first time. The changes of contraction frequency and contraction amplitude of myocardium after treatment with polydatin were determined by computer image analysis system. Results When 0.1 mmol·L-1 polydatin was added to the cultured cells, the contraction frequency of cardiomyocytes was significantly increased and the contraction amplitude was increased. When the digitalis drug Cedilan was added, the contraction amplitude of cardiomyocytes was increased and the beat frequency was also increased. However, the beating of cardiomyocytes in the control group with D Hanks did not change within 30 min. After administration of polydatin, the intracellular calcium fluorescence intensity of myocardial cells showed a progressive upward trend, rising to the initial 2 24% ± 2433%. Conclusion Polydatin may directly increase the contractility of cardiomyocytes by increasing the concentration of calcium ion in cardiomyocytes.
