目的观察“通利大肠”对慢性阻塞性肺疾病(COPD)模型大鼠气道黏液高分泌的影响,从黏液高分泌角度探讨COPD“从肠论治”的效应机制。方法采用气管注脂多糖加熏香烟联合造模方法建立COPD大鼠模型,随机分为正常组、模型组、治肺组、治肠组及肺肠同治组。正常组、模型组灌胃生理盐水,各给药组灌胃相应中药,连续14 d。实时荧光定量PCR法及免疫组化法检测支气管组织水通道蛋白5(AQP5)mRNA、黏蛋白5AC(MUC5AC)mRNA水平及蛋白表达。结果与正常组比较,模型组大鼠气道AQP5 mRNA水平及蛋白表达减少,MUC5AC mRNA水平及蛋白表达增加(P<0.01)。与模型组比较,治肠组AQP5 mRNA水平及蛋白表达增强,MUC5AC mRNA水平及蛋白表达减少(P<0.01)。与治肺组比较,肺肠同治组AQP5 mRNA水平及蛋白表达增强,MUC5AC mRNA水平及蛋白表达减少(P<0.01)。结论通利大肠,或在治肺的基础上增加通利大肠,均能降低MUC5AC mRNA水平及蛋白表达,增强AQP5mRNA水平及蛋白表达,从而抑制气道黏液高分泌,这可能是COPD“从肠论治”效应产生的作用环节之一。
Objective To investigate the effects of Tongliao on the airway mucus hypersecretion in rats with chronic obstructive pulmonary disease (COPD), and to explore the mechanism of COPD from the perspective of mucosal hypersecretion. Methods COPD model rats were established by tracheal lipopolysaccharide combined with cigarette smoking and were randomly divided into normal group, model group, treatment of lung group, treatment of intestine group and lung intestine. Normal group, model group, gavage with saline, each administration group gavage corresponding Chinese medicine, continuous 14 d. The mRNA and protein expression of AQP5 and MUC5AC in bronchial tissue were detected by real-time fluorescence quantitative PCR and immunohistochemistry. Results Compared with the normal group, the expression of AQP5 mRNA and protein in the airway decreased and the expression of MUC5AC mRNA and protein increased (P <0.01). Compared with the model group, the AQP5 mRNA and protein expression in the treatment group was significantly increased, while the MUC5AC mRNA and protein expression were decreased (P <0.01). Compared with the treatment group, the AQP5 mRNA and protein expression were significantly increased in the lung-intestine, and the expression of MUC5AC mRNA and protein were decreased (P <0.01). Conclusion Tonglida, or Tongliao large intestine based on the treatment of lung, can reduce MUC5AC mRNA and protein expression, enhance AQP5 mRNA and protein expression, thereby inhibiting airway mucus hypersecretion, which may be COPD On governance "one of the role of the link.
